NOVELTY – Producing a null mutant of the URA3 gene of Saccharomyces cerevisiae involves deleting complete copies of the gene using a yeast URA3 integrating cassette. The second copy of the yeast URA3 gene is deleted, and integrated with FGY010 cassette. The fragment homologous repair is used to produce second copy of the URA3 locus FGY020 genome. The Hph cassette containing hphMX4 the marker gene is withdrawn. The yeast genome FGYQ3G is integrated with the core cassette. The genetic markers kanMX4 and KiURA3 are withdrawn by using same fragment of repair. USE – Method for producing null mutant of the URA3 gene of Saccharomyces cerevisiae for metabolizing various sources to produce different industrial metabolites such as alcohol, glycerol, ethylene, propylene and biodiesel hydrocarbons, and for producing alcohol from canade-sugar (all claimed). ADVANTAGE – The method enables to produce a null mutant of the URA3 gene of Saccharomyces cerevisiae with good fermentation performance and excellent adaptation to the industrial conditions.
