Production process for violacein and deoxyviolacein is optimized by factorial planning and analysis of response surface for chromobacterium violaceum
NOVELTY – Production process for violacein and deoxyviolacein is optimized by factorial planning and analysis of response surface for chromobacterium violaceum and is based on statistical principles. USE – For the production of violacein and deoxyviolacein. ADVANTAGE – The process results in a 50 per cent increase in the production of crude violacein and a clear increase in cellular mass. DETAILED DESCRIPTION – Production process for violacein and deoxyviolacein is optimized by factorial planning and analysis of response surface for chromobacterium violaceum and is based on statistical principles. In the initial phase 11 factors are analyzed: source of carbon (glucose), yeast extract, methionine, vitamin B12, L-tryptophan, agitation, temperature, pH (phosphate tampon), peptone, inoculum and concentration of zinc sulfate. In a second phase, a second composed planning involved 5 factors: source of carbon (glucose), yeast extract, L-tryptophane, peptone and zinc sulfate. They were chosen based on the result of the previous factorial planning. In the final phase of optimization, three factors: source of carbon (glucose), yeast extract and L-tryptophane were studied, using a central composed planning of three factors. The average production of crude violacein in this planning was 0.36 g/L, which is 50 per cent greater than in the other models studied. The average of cellular mass for this planning was 16.59 g/L, around 2.2 times greater than in the other models studied.
